目的探讨室内二氧化碳(CO_2)体积分数对CO_2酶法试剂盒稳定性的影响。方法选用2个CO_2酶法试剂盒,采用2种方法进行试验:(1)试剂盒1在每天试验完成后封口置4℃冰箱保存;(2)试剂盒2模拟正常工作状态,持续敞口放置于仪器试剂仓(4℃)内直至全部试验完成。使用高值质控品、低值质控品、混合血清3种样本,分别在不同时间点使用2个试剂盒测定CO_2水平,同时测定室内CO_2体积分数,观察CO_2酶法试剂盒测定结果随室内CO_2体积分数变化的情况。3种样本的检测结果与室内CO_2体积分数的相关性采用Pearson相关分析。结果随着试剂盒装载时间的延长,使用2种试验方法测定高、低值质控品及混合血清CO_2结果的变化趋势与室内CO_2体积分数相同。相关性分析显示,高、低值质控品及混合血清使用2种试验方法测得的CO_2结果均与室内CO_2体积分数高度相关[试剂盒1(方法1):r值分别为0.722、0.853、0.790,P<0.01;试剂盒2(方法2):r值分别为0.689、0.875、0.801,P<0.01],与试剂盒装载时间均无相关性[试剂盒1(方法1):r值分别为-0.132、-0.108、-0.103,P>0.05;试剂盒2(方法2):r值分别为-0.124、-0.112、-0.111,P>0.05]。结论 CO_2酶法试剂盒稳定性受室内CO_2体积分数影响较大,实验室可通过换用封口试剂或通过减少人口密度、加大换气频率等方式避免室内CO_2体积分数产生较大波动,保证CO_2酶法检测结果的准确性。
Objective To investigate the effect of CO 2 volume fraction on the stability of CO_2 enzymatic kit. Methods Two CO2 enzymatic kits were selected and tested by two methods: (1) Kit 1 was sealed and stored in refrigerator at 4 ℃ after the completion of daily test; (2) Kit 2 simulated normal working condition and kept open for exposure In the instrument reagent warehouse (4 ℃) until all the tests are completed. CO 2 levels were determined by two kits at different time points, and the CO_2 volume fraction was determined at the same time. The results of CO_2 enzymatic kit assayed with the indoor CO_2 volume fraction changes. Pearson correlation analysis was used to test the correlation between the three kinds of samples and the indoor CO 2 volume fraction. Results With the extension of kit loading time, the trend of CO 2 in high and low quality control products and mixed serum determined by two test methods was the same as the indoor CO 2 volume fraction. Correlation analysis showed that CO 2 results measured by two test methods of high and low quality control products and mixed serum were highly correlated with indoor CO 2 volume fraction [kit 1 (method 1): r values were 0.722, 0.853, 0.790, P <0.01; kit 2 (method 2): r values were 0.689,0.875,0.801, P <0.01], and were not related to the kit loading time Was -0.132, -0.108, -0.103, P> 0.05; kit 2 (method 2): r values were -0.124, -0.112, -0.111, P> 0.05].
