目的观察诱导分化剂苯乙酸对胶质瘤细胞C6的增殖分化过程中癌基因C-myc的蛋白水平变化。方法[~3H]- TdR掺入法测细胞增殖率;免疫组化SP法检测C-myc基因蛋白水平的表达。结果应用苯乙酸后,细胞CPM值降低;C-myc 基因胞浆阳性表达显著下降。结论苯乙酸对胶质瘤细胞存在剂量依赖性抑制,作用机制可能为直接抑制胞浆中RNA合 成。这些作用与苯乙酸抑制C-myc基因对细胞恶性增殖的转录调节作用有关。
Objective To observe the changes of the protein level of oncogene C-myc in the differentiation and differentiation of C6 glioma cell induced by phenylacetate. Methods The [~ 3H] - TdR incorporation method was used to measure the cell proliferation rate. The expression of C - myc gene protein was detected by immunohistochemical SP method. Results After using phenylacetic acid, the cell CPM value decreased; the cytoplasm positive expression of C-myc gene was significantly decreased. Conclusion Phenylacetate inhibits the proliferation of glioma cells in a dose-dependent manner. The mechanism may be that it directly inhibits the synthesis of RNA in cytoplasm. These effects are related to the inhibitory effect of phenylacetate on the transcriptional regulation of C-myc on malignant proliferation of cells.
