目的 :研究急性白血病 (AL)中 p5 3、bcl- 2的表达特性及相关性 ,分析其表达与细胞周期的关系。方法 :4 9例 AL 患者为实验组 ,同期住院的骨髓象完全正常的非血液系恶性疾病患者 14例为对照组 ,取骨髓标本 ,分离单个核细胞 ,用特异性单克隆抗体标记 ,流式细胞仪检测突变型 p5 3及bcl- 2基因蛋白表达 ,其中有 2 6例患者同时用流式细胞仪测定了细胞周期。结果 :AL 组 p5 3、bcl- 2蛋白表达阳性率分别为 4 4 .9%、4 9.0 % ,显著高于对照组 ,AL L 组与 ANL L 组差异无显著性 ,M3组显著低于其他非 M3ANL L 组。突变型 p5 3蛋白与 bcl- 2蛋白表达呈正相关 ,二者在白血病的发病上有协同作用。白血病组比对照组细胞周期中 S+G2 / M期细胞数低 ,p5 3、bcl- 2表达与细胞周期无关。结论 :突变型 p5 3、bcl- 2有致白血病的作用 ,二者在白血病的发展中可能起一定协同作用。联合检测突变型 p5 3及 bcl- 2对预测白血病的预后有一定价值
Objective: To study the expression and correlation of p53 and bcl-2 in acute leukemia (AL), and to analyze the relationship between the expression and cell cycle. Methods: Forty-nine patients with AL were selected as the experimental group. In the same period, 14 patients with completely normal bone marrow in non-hematologic malignancies were selected as the control group. Bone marrow samples were taken and mononuclear cells were isolated. The cells were labeled with specific monoclonal antibody Cytometry was used to detect the expression of mutant p53 and bcl-2 genes. Twenty-six of the patients tested for cell cycle by flow cytometry. Results: The positive rates of p53 and bcl-2 in AL group were 44.9% and 44.0%, respectively, which were significantly higher than those in control group, but there was no significant difference between AL L group and ANL L group Non M3ANL L group. Mutant p5 3 protein and bcl-2 protein expression was positively correlated, both in the pathogenesis of leukemia has a synergistic effect. The cell number of S + G2 / M phase in leukemia group was lower than that in control group, and the expression of p5 3, bcl-2 was not related to cell cycle. CONCLUSION: Mutant p5 3 and bcl-2 have the effect of leukemia, both of which may play a synergistic role in the development of leukemia. The combined detection of mutant p5 3 and bcl-2 has some value in predicting the prognosis of leukemia
