目的 探讨建立胸腺因子 D(TFD) /重组白细胞介素 2 (r IL- 2 )高效诱导人脑胶质瘤浸润淋巴细胞(GIL )的可行性。 方法 按 TFD与 r IL - 2的不同组合 ,分组在体外对 GIL进行培养 ,分别用 MTT法、4h5 1 Cr释放法和间接免疫荧光法检测其增殖、杀伤活性及 r IL- 2受体的表达情况。 结果 TFD与 r IL- 2共同诱导 GIL,与对照组相比 ,其增殖活性及对自体瘤细胞的杀伤活性显著增强且持续时间更长 (P<0 .0 1)。 结论 应用 TFD/r IL- 2体外诱导方法 ,可望获取足量高活性的 GIL 用于临床脑胶质瘤的过继免疫冶疗。
Objective To explore the feasibility of establishing glioma factor D (TFD)/recombinant interleukin 2 (r IL-2) to efficiently induce human brain glioma infiltrating lymphocytes (GIL). Methods According to different combinations of TFD and r IL-2, GILs were cultured in vitro. The proliferation, killing activity and expression of rIL-2 receptors were detected by MTT method, 4h51 Cr release assay and indirect immunofluorescence assay, respectively. Happening. RESULTS: Both TFD and rIL-2 induced GIL. Compared with the control group, the proliferation activity and killing activity against autologous tumor cells were significantly enhanced and lasted longer (P < 0.01). Conclusion The use of TFD/r IL-2 in vitro induction method is expected to obtain sufficient and high activity of GIL for adoptive immunotherapy of clinical glioma.
