目的:阐明温阳益气、活血化瘀复方抗阿片类物质依赖戒断综合征、调节下丘脑一垂体轴功能紊乱的中枢机制;方法:建立Mor依赖和戒断模型,应用ISHH法、IS法观察了不同处理方法对与阿片类物质依赖关系密切的β-End前体POMC、对性激素起主要调节作用的GnRH的mRNA表达及其蛋白合成水平变化的影响;结果:Mor依赖和戒断大鼠下丘脑POMC、GnRH mRNA的表达显著降低,β-End、GnRH的蛋白合成明显减少,中药复方能显著提高其表达与合成水平;结论:中药复方增加β-End蛋白合成、提高POMC mRNA表达,是其抑制戒断症状的中枢途径之一;增加GnRH mRNA表达和蛋白合成,是其调节紊乱性激素水平、减轻性器官损害的上行机制。
Objective: To elucidate the central mechanism of Wenyangyiqi and Huoxuehuayu compound anti-opioid withdrawal syndrome and regulate hypothalamus-pituitary axis dysfunction. Methods: Establish Mor dependency and withdrawal models, and observe with ISHH method and IS method The effect of different treatments on the mRNA expression and GnRH expression of GnRH, which is the main regulator of sex hormones, is closely related to the opioid dependence of POMC, and the changes of the protein synthesis levels; Results: Mor dependence and withdrawal in rat hypothalamus The expressions of POMC and GnRH mRNA were significantly decreased, and the protein synthesis of β-End and GnRH was significantly reduced. The Chinese herbal compound can significantly increase the expression and synthesis level; Conclusion: Chinese herbal compound increases β-End protein synthesis and increases the expression of POMC mRNA, which inhibits it. One of the central pathways for withdrawal symptoms; increase of GnRH mRNA expression and protein synthesis is an upward mechanism for regulating the level of disordered sex hormones and alleviating sexual organ damage.
