目的:观察用小鼠和人不同的精子抗原免疫雌小鼠对妊娠影响。方法:用小鼠2种和人4种精子抗原免疫雌小鼠,采用酶标法检测血清抗精子抗体(AsAb),观察各组小鼠妊娠率及胎仔数。结果:随着主动免疫的进行,小鼠血清AsAb显著升高(P<0.01)。精子抗原免疫小鼠8周后,小鼠精子混合抗原免疫小鼠效果最好,妊娠率为零,DTT提取的人精子膜抗原和人全精子抗原免疫小鼠效果较好,妊娠率为16.7%,显著低于正常对照组的63.6%(P<0.01或P<0.05)。结论:在人精子抗原中,DTT提取的人精子膜抗原和人全精子抗原免疫雌小鼠效果最好。在小鼠精子抗原中,DTT、NP-40、Triton-100提取的小鼠精子膜混合抗原免疫雌小鼠效果最好。
OBJECTIVE: To observe the effects of different immunization of mice and human sperm antigen on pregnancy in mice. Methods: Female mice were immunized with 2 kinds of mouse and 4 kinds of human sperm antigens. The serum anti-sperm antibody (AsAb) was detected by enzyme-linked immunosorbent assay (ELISA). The pregnancy rate and number of litter size in each group were observed. Results: Serum AsAb was significantly increased with the progress of active immunization (P <0.01). After 8 weeks of immunization with sperm antigen, mice immunized with sperm mixed antigen had the best effect and pregnancy rate was zero. The results of DTT-extracted human sperm membrane antigen and human whole sperm antigen immunized mice were better, the pregnancy rate was 16.7% , Significantly lower than 63.6% of the normal control group (P <0.01 or P <0.05). CONCLUSION: Among human sperm antigens, DTT-extracted human sperm membrane antigen and human whole sperm antigen are the best immunized female mice. Among the mouse sperm antigens, mice were best immunized with DTT, NP-40 and Triton-100 mouse sperm membrane mixed antigen.
