目的观察石菖蒲水提取成分对大鼠脑皮质NMDA、M受体与其配体结合的影响。方法提取脑皮质受体后,加入相应的石菖蒲提取成分,运用放射配基结合分析法检测NMDA、M受体与其相应配体的的结合。结果石菖蒲挥发油、去油水煎液、β-细辛醚均能拮抗NMDA受体与3H-MK 801的结合,但对M受体与3H-QNB的结合无影响。结论石菖蒲水提成分能拮抗大鼠脑皮质NMDA受体与其配体的结合,该作用可能是其醒脑开窍、治疗脑损伤的重要药理机制之一。
Objective To observe the effects of water extract of Acorus calamus on the binding of NMDA and M receptors to the cerebral cortex in rats. Methods After extraction of cerebral cortex receptors, the corresponding Acorus gramineus extracts were extracted, and the binding of NMDA and M receptors to their corresponding ligands was detected by radioligand binding assay. Results Acorus gramineus volatile oil, oil decoction and β-asarone both antagonized the binding of NMDA receptor to 3H-MK 801, but had no effect on the binding of M receptor to 3H-QNB. Conclusion The water extract of Acorus gramineus can antagonize the binding of NMDA receptor and its ligand in rat cerebral cortex, which may be one of the important pharmacological mechanisms for the brain to resuscitate and treat brain damage.
