目的观察探讨大鼠骨骼肌挫伤愈合过程中p-CB1R的表达规律与损伤时间的相关性。方法健康成年雄性SD大鼠50只,体重220~250g,随机分为10组,每组5只,其中9组为实验组,1组为正常对照组。应用免疫组织化学染色和Western blot技术,检测50例大鼠骨骼肌挫伤后各时间段p-CB1R的表达情况。结果正常对照组大鼠骨骼肌未见p-CB1R表达;挫伤后p-CB1R阳性表达主要在单核细胞和成纤维细胞中,各时间段大鼠骨骼肌均未见阳性反应。伤后3~24h,阳性细胞以单核细胞为主;3~5d,阳性细胞以单核细胞和成纤维细胞为主;7~10d,阳性反应主要见于成纤维细胞,并于7d达到高峰;14d,p-CB1R阳性表达量有所下降。结论骨骼肌挫伤愈合过程中p-CB1R在单核细胞和成纤维细胞中的变化具有时序性,可望作为法医学推断骨骼肌挫伤时间的参考指标之一。
Objective To investigate the correlation between the expression of p-CB1R and the injury time during the contusion healing of skeletal muscle in rats. Methods Fifty healthy adult male Sprague-Dawley rats weighing 220-250 g were randomly divided into 10 groups with 5 rats in each group, of which 9 were experimental group and 1 group was normal control group. Immunohistochemical staining and Western blot were used to detect the expression of p-CB1R at each time point after the contusion of skeletal muscle of 50 rats. Results No expression of p-CB1R was found in skeletal muscle of rats in normal control group. The expression of p-CB1R in contusion group was mainly in monocytes and fibroblasts, and no positive reaction was found in skeletal muscle of rats at each time. At 3 ~ 5 days after injury, the majority of positive cells were monocytes; the positive cells were mainly mononuclear cells and fibroblasts after 3 ~ 5 days; the positive reaction was mainly seen in fibroblasts from 7 to 10 days after injury, reaching the peak at 7 days; 14d, p-CB1R positive expression decreased. Conclusion The change of p-CB1R in mononuclear cells and fibroblasts during the process of skeletal muscle contusion healing is sequential, which is expected to be used as a reference index for judging the contusion time of skeletal muscle in forensic science.
