21.应用DNA指纹图法对妊娠初期流产的研究(高桥完明他) 作为流产的原因之一,历来认为是由于胎儿染色体异常所致,至今染色体分析法仍为检查方法的主流。1985年Jeffreys等创立了DNA指纹图(DNA fingerprint)法,现逐渐认识到,本法在个体判别、亲子鉴定等方面极为有用。本文报告了利用DNA指纹图法对流产的遗传基因进行分析的尝试。分离妊娠初期流产绒毛及双亲静脉血中的有核细胞,抽提DNA。将DNA用限制性内切酶切断后,在0.9%琼脂凝胶上进行电泳,然后在硝酸纤维滤纸上进行Southern转移。用~(32)P标记的短的随体DNA作为探针进行杂交,比较流产绒毛及双亲

The application of DNA fingerprinting method for early pregnancy miscarriage (Takahashi completed his) as one of the reasons for abortion, has always been that due to fetal chromosomal abnormalities, so far the main method of chromosomal analysis. In 1985, Jeffreys founded the DNA fingerprint method. Now it is gradually realized that this method is extremely useful in individual discrimination and paternity testing. This paper reports an attempt to analyze the genetic profile of abortion using DNA fingerprinting. Isolation of early pregnancy abortion villus and parenchyma nucleated cells, DNA extraction. The DNA was cut with a restriction enzyme, electrophoresed on a 0.9% agarose gel, and then subjected to Southern blotting on a nitrocellulose filter. The short satellite DNA labeled with ~ (32) P was used as a probe for hybridization, and the miscarried fluff and parents were compared
