目的:研究青蒿琥酯对皮肤癌细胞核形态变化影响及Ag NOR颗粒形态影响。方法:购买人表皮鳞癌细胞株A431细胞株,随机分为两组,常规培养后,200μmol/L的青蒿琥酯溶液(经10%胎牛血清稀释)1 m L;对照组加入等量的10%胎牛血清,培养48 h后制作切片;采用HE染色和银染色处理切片。通过彩色病理图像分析系统观察两组细胞核形态、细胞结构、Ag NOR颗粒形态以及Ag NOR颗粒分型。结果:1实验后实验组细胞细胞核的面积(P)、周长(At)、最大直径(D),与对照组比较均较低,差异有统计学意义(P<0.05);2实验后实验组细胞细胞核体密度(Vv)、表面积密度(Sv)、平均体积(V)、平均表面积(S)以及三维形状因子(Rsv),与对照组比较均较低(P<0.05),实验组核数密度(Nv)明显高于对照组(P<0.05);3实验后实验组Ag NOR颗粒的直径大于对照组(P<0.05);实验组Ag NOR颗粒的截面数、面积以及其余和面积的比值均低于对照组(P<0.05);4实验后实验组单一型、核仁内型Ag NOR颗粒所占的百分比高于对照组,而聚集型、弥散型Ag NOR颗粒所占的百分比低于对照组,差异有统计学意义(χ~2=6.313,P<0.05)。结论:青蒿琥酯能够使皮肤癌细胞核形态及Ag NOR颗粒形态趋于规则,抑制肿瘤细胞增殖,对临床具有指导意义,值得临床推广。
Objective: To study the effect of artesunate on the nuclear morphology of skin cancer cells and the morphological changes of Ag NOR particles. Methods: Human epidermal growth factor (SCC) cell line A431 was purchased and randomly divided into two groups. The number and area of Ag NOR particles in the experimental group were significantly higher than those in the control group (P <0.05) .4 After experiment, the percentages of single type and nucleolar type Ag NOR particles in the experimental group were higher than those in the control group, while the percentage of aggregated and diffuse type Ag NOR particles was low In the control group, the difference was statistically significant (χ ~ 2 = 6.313, P <0.05). CONCLUSION: Artesunate can make the nuclear morphology of skin cancer cells and the morphology of Ag-NOR particles regular and inhibit the proliferation of tumor cells, which is of clinical significance and worthy of clinical promotion.
