目的研究老年时期规律运动和蜕皮素诱导基因表达量降低对老龄果蝇心脏功能的影响,同时对果蝇老年时期运动抗心脏衰老的效果进行评价。方法安静组、干扰组、运动组和干扰运动组前6周均保存在标准培养箱内,第7周开始运动时,保存在30℃、其他条件与之前相同的培养箱内。第44天龄开始,运动组和干扰运动组果蝇采用运动训练平台装置进行规律性训练,环境温度为30℃,每天运动2.5 h,连续运动5天。全部运动结束后的第2天解剖果蝇,利用M-mode测试系统检测其心功能。结果运动结束后,收缩直径安静组小于干扰组(P<0.05),运动组小于干扰运动组(P<0.05);舒张直径安静组小于运动组(P<0.01);纤维性震颤安静组低于干扰组(P<0.05),干扰运动组低于干扰组(P<0.05),运动组低于安静组(P<0.05);射血分数安静组高于干扰组(P<0.01),干扰运动组高于干扰组(P<0.01)。结论蜕皮素诱导基因表达量的降低使果蝇心脏收缩力减弱,纤维性震颤的发生率升高,射血分数减少,而运动能够减少老龄果蝇心脏纤维性震颤的发生率,增加射血分数,有效提高果蝇心肌收缩力。
Objective To study the effect of regular exercise and the expression of ecdysone-induced genes on heart function in aged Drosophila melanogaster during old age, and to evaluate the effect of anti-heart aging on Drosophila in old age. Methods The quiet group, the disturbance group, the exercise group and the disturbance exercise group were kept in the standard incubator for the first 6 weeks. At the beginning of the 7th week, the exercise was kept at 30 ° C and the other conditions were the same in the same incubator. Conclusion The decrease of ecdysone-induced gene expression leads to decreased cardiac contractility, increased fibrillation and decreased ejection fraction, while exercise can reduce the incidence of cardiac fibrillation and increase the ejection fraction , Effectively improve Drosophila myocardial contractility.
