目的:获得特异的鼠抗人尿激酶型纤溶酶原激活剂的单克隆抗体,以建立特异、灵敏、准确的癌症临床诊断方法。方法:以纯化的尿激酶型纤溶酶原激活剂(uPA)为抗原免疫Balb/c小鼠,制备单克隆抗体。结果:利用小鼠杂交瘤技术,建立4 株分泌uPA 抗体的杂交瘤细胞系6G9、6G10、5E12、6H2,经鉴定其抗体类别均为IgG,它们均能特异识别uPA,与其它因子无交叉反应。结论:提示癌瘤组织中uPA 含量对临床诊断及预测转移的可能性具有临床意义
OBJECTIVE: To obtain monoclonal antibodies specific to human anti-human urokinase plasminogen activator in order to establish a specific, sensitive and accurate method for clinical diagnosis of cancer. Methods: Monoclonal antibodies were prepared by immunizing Balb / c mice with purified urokinase plasminogen activator (uPA). Results: Four hybridoma cell lines secreting uPA antibody were established by hybridoma technology of mouse, 6G9, 6G10, 5E12 and 6H2. All the antibodies were identified as IgG. All of them could recognize uPA specifically and did not cross-react with other factors . Conclusion: It is indicated that the uPA content in cancer tissue is of clinical significance for clinical diagnosis and prediction of metastasis
