目的:通过HPLC测定浸渍不同温度、不同时间条件下大黄黄连泻心汤中大黄素、小檗碱、黄芩苷含量的差异,初步揭示《伤寒论》麻沸汤浸渍条件。方法:设定100℃、85℃、70℃不同浸渍温度,均分别浸渍5 min、15 min、48min,获得9组大黄黄连泻心汤浸渍液。运用高效液相色谱测定。色谱柱为XBridge~(TM)C-_(18)(250 mm×4.6 mm,5μm),流动相为甲醇-0.1%磷酸梯度洗脱,检测波长为280 nm,流速为0.8 m L·min~(-1),柱温为30℃,进样量10μL。理论塔板数以黄芩苷计不低于1500。结果:小檗碱在100℃热水浸渍48 min的条件下浓度最大,黄芩苷在85℃热水浸渍48 min的时候浓度最大,大黄素在70℃水温浸渍15 min的条件下浓度最高。结论:大黄黄连泻心汤麻沸汤浸渍的方法可能为85℃浸渍15 min。
OBJECTIVE: To determine the content of emodin, berberine and baicalin in rhubarb Huangxie Xiexin decoction by immersing in different temperature and time, and to reveal the impregnation conditions of “Maosang Tang” in “Treatise on Febrile Diseases”. Methods: Different immersion temperatures of 100 ℃, 85 ℃ and 70 ℃ were set and immersed for 5 min, 15 min and 48 min, respectively. Nine groups of Rhubarb Huangxiexinsang Decoction were obtained. Using high performance liquid chromatography. The mobile phase consisted of a gradient of methanol-0.1% phosphoric acid with a gradient of 280 nm at a flow rate of 0.8 m L · min ~ (-1). The column temperature was 30 ℃ and the injection volume was 10μL. The number of theoretical plates to baicalin not less than 1500. Results: Berberine had the highest concentration under the conditions of 100 ℃ hot water immersion for 48 min. Baicalin had the highest concentration when heated at 85 ℃ for 48 min. Emodin was the highest concentration when immersed at 70 ℃ for 15 min. Conclusion: The method of immersing Rhubarb Huangxiexin Decoction may be 15 min at 85 ℃.
