目的:探讨驱虫斑鸠菊对人 A375黑素瘤细胞株分泌酪氨酸酶活性、黑素合成以及细胞内酪氨酸酶基因表达水平的影响。方法:采用酶学方法研究驱虫斑鸠菊对酪氨酸酶活性的影响;采用475 nm 比色法测定黑素含量;采用 TRIzol 试剂盒提取总 RNA,再用半定量逆转录聚合酶链反应扩增 cDNA,检测酪氨酸酶基因表达水平。结果:驱虫斑鸠菊可以增强 A375人黑素瘤细胞酪氨酸酶活性和黑色素合成能力,还可增强酪氨酸酶基因表达。结论:驱虫斑鸠菊可能从基因水平增强酪氨酸酶活性,促进黑素合成。
Objective: To investigate the effect of versicolor anthelmintica on the secretion of tyrosinase activity, melanin synthesis, and intracellular tyrosinase gene expression in human A375 melanoma cell lines. METHODS: Enzymatic methods were used to study the effect of Vernonia anthelmintica on tyrosinase activity; melanin content was determined by 475 nm colorimetric method; total RNA was extracted by TRIzol kit and semi-quantitative reverse transcriptase polymerase chain reaction amplification was used. Add cDNA to detect tyrosinase gene expression levels. Results: Vernonia anthelmintica could enhance the activity of tyrosinase and melanin synthesis in A375 human melanoma cells, and increase the expression of tyrosinase gene. Conclusion: Vernonia anthelmintica may increase tyrosinase activity from gene level and promote melanin synthesis.
