目的:探讨肿瘤坏死因子-α(TNF-α)SNP308G/A多态性与头颈癌风险的关系。方法:检索文献并按要求提取相关信息,纳入TNF-αSNP308G/A与头颈癌风险的病例-对照研究,对研究结果进行Meta分析。结果:共纳入10个病例-对照研究,包括1833例头颈癌患者和2542例对照。杂合子模型(AA vs.GA)分析表明,与GA基因型个体相比较,AA基因型个体与发生头颈癌的高风险存在显著关联性(AA vs.GA:OR=2.48,95%CI,1.48-4.16,P<0.001)。亚组分析亦表明,有显著相关性的结果主要体现在杂合子模型中(AA vs.GA),主要集中在口腔癌,主要发生在亚洲人群中和医院来源的对照组分析中。所纳入研究未发现显著的发表偏倚。结论:TNF-α基因SNP308多态性与头颈癌易感性显著相关,可能是发生头颈癌的高风险因素。
Objective: To investigate the relationship between SNP308G / A polymorphism of tumor necrosis factor-α (TNF-α) and the risk of head and neck cancer. METHODS: The literature was retrieved and relevant information was extracted as required to include a case-control study of TNF-α SNP308G / A and head and neck cancer risk. Meta-analysis was performed on the results. RESULTS: Ten case-control studies were included, including 1833 head and neck cancer patients and 2542 controls. AA vs. GA analysis showed that there was a significant association between individuals with AA genotype and high risk of head and neck cancer compared with individuals with GA genotype (AA vs. GA: OR = 2.48, 95% CI, 1.48 -4.16, P <0.001). Subgroup analyzes also showed that significant correlations were mainly found in heterozygous models (AA vs. GA), mainly in oral cancers, mainly in Asian and hospital-derived controls. The included studies did not find significant publication bias. Conclusion: SNP308 polymorphism of TNF-α gene is significantly associated with susceptibility to head and neck cancer and may be a high risk factor for head and neck cancer.
