目的研究油酸(oleic acid,OA)及其反式异构体反油酸(elaidic acid,EA)对人成熟脂肪细胞脂联素mRNA表达的影响。方法原代培养人脂肪组织来源的间质干细胞(human adipose tissue-derived stromal cells,hATSCs),诱导分化为成熟脂肪细胞,分别暴露于0(对照)、1、5、25μmol/L的OA和EA溶液72 h。采用实时定量PCR(real time PCR)方法检测脂联素mRNA的相对表达水平。结果与对照组比较,当人成熟脂肪细胞暴露于5、25μmol/L OA时,脂联素mRNA表达水平降低,差异有统计学意义(P<0.05);当人成熟脂肪细胞暴露于1、5、25μmol/L EA时,脂联素mRNA表达水平升高,差异有统计学意义(P<0.05)。且人成熟脂肪细胞脂联素mRNA表达水平随着OA染毒浓度的升高而下降,随着EA染毒浓度的升高而上升。与相同暴露剂量的OA组比较,EA暴露组人成熟脂肪细胞脂联素mRNA表达水平均高,差异有统计学意义(P<0.05)。结论 OA具有抑制人成熟脂肪细胞脂联素mRNA表达的作用,而EA具有增强作用。
Objective To investigate the effect of oleic acid (OA) and its elaidic acid (EA) on the mRNA expression of adiponectin in human mature adipocytes. Methods Primary human adipose tissue-derived stromal cells (hATSCs) were cultured and differentiated into mature adipocytes. The cells were exposed to 0 (control), 1, 5, 25μmol / L of OA and EA Solution 72 h. The real-time PCR method was used to detect the relative expression of adiponectin mRNA. Results Compared with the control group, the expression of adiponectin mRNA decreased when mature adipocytes were exposed to 5,25 μmol / L OA (P <0.05). When mature adipocytes were exposed to 1,5 , And 25μmol / L EA, adiponectin mRNA expression increased, the difference was statistically significant (P <0.05). Adiponectin mRNA expression in mature human adipocytes decreased with the increase of OA concentration and increased with the increase of EA concentration. Compared with OA group with the same exposure dose, adiponectin mRNA expression level in mature adipocytes of EA-exposed group was significantly higher than that of OA group (P <0.05). Conclusion OA has an inhibitory effect on adiponectin mRNA expression in human mature adipocytes, whereas EA has an enhancing effect.
