目的探讨CCR5拮抗剂Maraviroc对人唾液腺腺样囊性癌(SACC)细胞运动、趋化、侵袭及增殖的影响。方法采用体外划痕、趋化、侵袭及增殖实验,观察Maraviroc对SACC-83和SACC-LM细胞的运动、趋化、侵袭及增殖能力的影响。结果与对照组比较,加入Maraviroc预处理的实验组细胞划痕模型愈合速度明显降低;趋化和侵袭实验中,穿越多聚碳酸酯膜的细胞数明显减少;增殖实验结果显示可明显抑制SACC-83和SACC-LM细胞增殖。结论 Maraviroc可能是一种新的靶向抗肿瘤药物,小分子剂量的临床应用可能会抑制SACC的发展。
Objective To investigate the effects of CCR5 antagonist Maraviroc on the motility, chemotaxis, invasion and proliferation of human salivary adenoid cystic carcinoma (SACC) cells. Methods Scratch, chemotaxis, invasion and proliferation assays were performed in vitro to observe the effects of Maraviroc on the motility, chemotaxis, invasion and proliferation of SACC-83 and SACC-LM cells. Results Compared with the control group, the healing rate of the cell scratch model in Maraviroc pretreatment group was significantly reduced. In the chemotaxis and invasion experiments, the number of cells crossing the polycarbonate membrane was significantly reduced. The results of proliferation assay showed that SACC- 83 and SACC-LM cell proliferation. Conclusion Maraviroc may be a new targeted anti-tumor drug. The clinical application of small molecule dose may inhibit the development of SACC.
