目的观察香菇多糖(lentinan,Lent)对约氏疟原虫(Plasmodium yoelii 17XL,Py17XL)感染小鼠树突状细胞(dendritic cells,DCs)的免疫调节作用。方法 120只BALB/c小鼠随机分为实验组和对照组,腹腔接种1×106Py17XL寄生的红细胞后,实验组用香菇多糖腹腔注射处理,对照组仅采用PBS腹腔注射处理,分别统计小鼠的平均生存时间和原虫血症水平;分离小鼠脾脏DCs细胞,用Western blot检测其IL-2、IL-10、IL-12、TGF-β、CD86和TLR-9蛋白表达水平。结果实验组和对照组小鼠平均生存时间为(8.35±2.55)d和(5.30±1.26)d,差异有统计学意义(P<0.05)。实验组脾脏DCs细胞IL-2、IL-12、TGF-β、CD86和TLR-9蛋白表达水平显著高于对照组(P<0.05),IL-10蛋白表达水平显著低于对照组(P<0.05)。结论香菇多糖具有抗疟作用,能通过上调IL-2、IL-12、TGF-β、CD86和TLR-9等细胞因子水平促使DCs成熟和加强抗原递呈作用。
Objective To investigate the immunomodulatory effects of lentinan (Lent) on dendritic cells (DCs) infected with Plasmodium yoelii 17XL (Py17XL). Methods 120 BALB / c mice were randomly divided into experimental group and control group. After intraperitoneal inoculation of 1 × 106Py17XL parasitic erythrocytes, the experimental group received intraperitoneal injection of lentinan and the control group received intraperitoneal injection only of PBS Mean survival time and parasitemia were detected. The splenic DCs were isolated and the protein expressions of IL-2, IL-10, IL-12, TGF-β, CD86 and TLR-9 were detected by Western blot. Results The average survival time of mice in experimental group and control group was (8.35 ± 2.55) d and (5.30 ± 1.26) d, respectively, with statistical significance (P <0.05). The protein expression levels of IL-2, IL-12, TGF-β, CD86 and TLR-9 in splenic DCs in experimental group were significantly higher than those in control group (P <0.05) 0.05). Conclusion Lentinan possesses the anti-malarial effect and can promote the maturation of DCs and enhance the antigen presentation by up-regulating the levels of cytokines such as IL-2, IL-12, TGF-β, CD86 and TLR-9.
