以树兰(Epidendrum)为材料,分别研究了NaClO和HgCl2对树兰侧芽、茎段和花梗3种外植体的消毒效果,探讨树兰3种外植体的有效消毒方法。试验结果表明:(1)3种外植体中最易消毒的为花梗,其次是侧芽,最难消毒的为茎段;(2)HgCl2对3种外植体的消毒效果优于NaClO;(3)3种外植体最适宜的消毒方法分别为:花梗0.1%HgCl2消毒8 min,侧芽0.1%HgCl2消毒4 min,茎段0.1%HgCl2消毒10 min,其存活率分别为90.9%、42.3%和30.9%。本研究结果将为树兰外植体途径的组培再生提供理论依据和技术指导。
Taking Epidendrum as material, the disinfection effects of NaClO and HgCl2 on the explants of lateral buds, stem segments and pedicels of Eucalyptus globulus were studied respectively, and the effective disinfection methods of three explants of Eucalyptus species were discussed. The results showed that: (1) The most easily disinfected species of three explants were pedicel, followed by lateral buds, and the most difficult to sterilize was stem segments; (2) HgCl2 had better disinfection effect than NaClO on three explants; ( 3) The best methods for disinfection of three kinds of explants were: disinfection of pedicel 0.1% HgCl2 for 8 min, lateral bud 0.1% HgCl2 disinfection for 4 min, sterilized 0.1% HgCl2 for 10 min, the survival rates were 90.9%, 42.3% And 30.9%. The results of this study will provide theoretical basis and technical guidance for the tissue culture regeneration of the explants of tree orchid.
