目的探讨Patched1(Ptch1)、融合阻抑蛋白Su(Fu)基因在胰腺癌中的表达及其与胰腺癌发生、发展的关系。方法采用RT-PCR、Western blot法检测56例胰腺癌组织中Ptch1、Su(Fu)基因和蛋白的表达情况,并分析其与胰腺癌临床病理参数之间的关系。结果 RT-PCR检测结果显示,Ptch1、Su(Fu)m RNA在胰腺癌中相对表达量分别为0.413±0.045、0.376±0.053,在癌旁组织中分别为0.203±0.013、0.214±0.062(P<0.05)。Western blot法检测结果显示,Ptch1、Su(Fu)蛋白在胰腺癌中的相对表达量分别为0.724±0.021、0.625±0.075,在胰腺癌旁组织中分别为0.516±0.098、0.413±0.051。胰腺癌组织中Ptch1、Su(Fu)m RNA与蛋白表达均高于癌旁组织。胰腺癌组织中Ptch1、Su(Fu)m RNA与蛋白表达与胰腺癌的分化程度和TNM分期明显相关(P<0.05),与患者年龄、性别、肿瘤直径、侵犯血管和神经无明显相关(P>0.05)。结论胰腺癌组织中Ptch1、Su(Fu)蛋白呈高表达,Ptch1、Su(Fu)蛋白的高表达可能与胰腺癌的发生及发展有关。
Objective To investigate the expression of Patched1 (Ptch1) and Su fusion gene in pancreatic cancer and its relationship with the occurrence and development of pancreatic cancer. Methods The expressions of Ptch1 and Su (Fu) genes and proteins in 56 cases of pancreatic cancer were detected by RT-PCR and Western blot. The relationship between them and the clinicopathological parameters of pancreatic cancer were analyzed. Results The RT-PCR results showed that the relative expression levels of Ptch1 and Su (Fu) m RNA in pancreatic cancer were 0.413 ± 0.045 and 0.376 ± 0.053, respectively, and were 0.203 ± 0.013 and 0.214 ± 0.062 in paracancerous tissues (P < 0.05). The results of Western blot showed that the relative expression levels of Ptch1 and Su (Fu) proteins in pancreatic cancer were 0.724 ± 0.021 and 0.625 ± 0.075, respectively, and were 0.516 ± 0.098 and 0.413 ± 0.051 in pancreatic cancer tissues. The expressions of Ptch1, Su (Fu) m RNA and protein in pancreatic cancer tissues were higher than those in paracancerous tissues. The expressions of Ptch1, Su (Fu) m RNA and protein in pancreatic cancer were significantly correlated with the differentiation degree and TNM stage of pancreatic cancer (P <0.05), but not with the age, sex, tumor diameter and invasion of blood vessels and nerves > 0.05). Conclusion The expressions of Ptch1 and Su (Fu) proteins in pancreatic cancer tissues are highly expressed. The high expression of Ptch1 and Su (Fu) proteins may be related to the occurrence and development of pancreatic cancer.
