目的 研究汉族人群中 139个微卫星位点的杂合度和多态信息量 (polymorphisminformation content,PIC)。方法 应用多重 PCR方法对 139个位点进行扩增。聚丙烯酰胺凝胶电泳分离 ,Gene Scan TM、Genotyper TM软件进行基因分型。结果 这些微卫星位点的杂合度为 0 .35~ 0 .89,其中 88%的杂合度 >0 .6 0 ,PIC为 0 .32~ 0 .88,94%位点的 PIC>0 .5 0。结论 微卫星位点有较高的多态信息量 ,在不同群体间存在一定的差异 ,在遗传分析中有广泛的应用价值。
Objective To study the heterozygosity and polymorphism information content (PIC) of 139 microsatellite loci in Han population. Methods Multiplex PCR was used to amplify 139 loci. Polyacrylamide gel electrophoresis, Gene Scan (TM), Genotyper (TM) software for genotyping. Results The heterozygosity of these microsatellite loci was 0.35-0.89, of which 88% had heterozygosity> 0.60, PIC was 0.32-0.88, PIC at 0% and 94% 0. Conclusion Microsatellite loci have higher polymorphic information content, and there are some differences among different populations. They have extensive application value in genetic analysis.
