具有抗氧化作用的维生素对高铁负荷大鼠抗氧化能力的影响

为研究具有抗氧化作用的维生素 (维生素C ,维生素E和β 胡萝卜素 )对高铁负荷大鼠脂质过氧化的影响 ,将 4 8只SD雄性大鼠随机分成 4组 ,实验组在每日饲以高铁饲料 (2 5 %碳酰铁 )的基础上 ,每日分别给予维生素C 10 0mg kgBW ,维生素E 5 0mg kgBW和β 胡萝卜素 (β C) 5 0mg kgBW ,每日对照组饮双蒸水 10mL kgBW ,实验期为 2 8d。实验结束时 ,测定血清和肝匀浆中铁、MDA、GSH含量 ,SOD活性和总抗氧化能力 (T AOC) ,肝匀浆中GSH Px活性。结果显示添加维生素C后 ,除肝匀浆中铁含量显著增加 (P <0 .0 5 )外 ,各项指标均与对照组无显著性差异。添加维生素E后 ,肝匀浆中铁含量显著提高 (P <0 .0 1) ,SOD和GSH Px活力显著下降 (P <0 .0 1)。添加 β 胡萝卜素后 ,肝匀浆中MDA含量显著降低 (P <0 .0 1) ,T AOC显著提高 (P <0 .0 5 ) ,GSH含量、SOD和GSH Px活性显著降低。推测在高铁负荷模型中 β 胡萝卜素可能通过非酶促体系发挥其抗氧化作用 ,维生素E、维生素C均可促进铁在肝脏的蓄积 ,对高铁负荷动物模型的抗氧化体系无保护作用 ,维生素E可能还有削弱作用 In order to study the effects of vitamins with antioxidant effects (vitamin C, vitamin E and β-carotene) on lipid peroxidation in rats with high iron overload, 48 SD male rats were randomly divided into 4 groups. On the basis of high-speed rail (25% ferricyanide), vitamin C 10 0 mg kg BW, vitamin E 50 mg kg BW, and β carotene 50 mg kg BW were given daily. The control group received double distilled water 10mL kgBW, the experimental period was 28 days. At the end of the experiment, the content of iron, MDA and GSH, the activity of SOD and total antioxidant capacity (T AOC) in serum and liver homogenate and the activity of GSH Px in liver homogenate were measured. The results showed that addition of vitamin C, in addition to liver iron content increased significantly (P <0. 05), the indicators were no significant difference with the control group. After adding vitamin E, the content of iron in liver homogenate increased significantly (P <0.01), the activities of SOD and GSH Px decreased significantly (P <0.01). After addition of β-carotene, the content of MDA in liver homogenate decreased significantly (P <0.01), T AOC increased significantly (P <0.05), GSH content, SOD and GSH Px activity decreased significantly. It is speculated that β-carotene may exert its antioxidant effect through non-enzymatic system in high-iron load model. Vitamin E and vitamin C may promote the accumulation of iron in the liver and have no protective effect on the antioxidant system of high-iron-loaded animal model.
