目的:建立无需衍生-超高效液相色谱法快速测定食品中黄曲霉毒素B1、B2、G1和G2的方法。方法:样品用甲醇水提取,提取液经黄曲霉毒素免疫亲和柱净化后,直接经C18色谱柱分离、荧光检测器检测。结果:4种黄曲霉毒素完全分离,AFG2、AFB2在0.010μg/L~2.00μg/L、AFB1在0.04μg/L~8.00μg/L、AFG1在0.10μg/L~20.0μg/L质量浓度范围内具有良好线性关系;最低检出浓度分别为0.04μg/kg(AFB1)、0.10μg/kg(AFG1)、0.01μg/kg(AFB2、AFG2);回收率为78.5%~110.5%,相对标准偏差(RSD)为2.46%~8.81%。结论:该方法简单、快速、灵敏,适用于食品中4种黄曲霉毒素含量的同时测定。
OBJECTIVE: To establish a rapid method for the determination of aflatoxins B1, B2, G1 and G2 in food without derivatization-ultra performance liquid chromatography. Methods: The sample was extracted with methanol water. The extract was purified by aflatoximin immunoaffinity column and directly separated by C18 column and detected by fluorescence detector. Results: Four kinds of aflatoxins were completely separated. The concentrations of AFG2 and AFB2 were 0.010μg / L ~ 2.00μg / L, AFB1 was 0.04μg / L ~ 8.00μg / L, AFG1 was 0.10μg / L ~ 20.0μg / L, (AFB1), 0.10μg / kg (AFG1) and 0.01μg / kg (AFB2, AFG2) respectively. The recoveries were 78.5% -110.5% with relative standard deviations (RSD) of 2.46% ~ 8.81%. Conclusion: The method is simple, rapid and sensitive and suitable for the simultaneous determination of four aflatoxins in food.
