目的:观察降脂益肝冲剂对非酒精性脂肪肝(NAFLD)大鼠肝脏肝型脂肪酸结合蛋白(L-FABP)mRNA表达的影响。方法:60只雄性Wistar大鼠随机分为6组(每组10只):正常组(N1,N2)给予普通饲料喂养;模型组(M1,M2)和治疗组(D1,D2)给予高脂饮食喂养;D1组在给予高脂饮食的基础上于0周时即给予降脂益肝冲剂,D2组于8周时给予降脂益肝冲剂,同时模型组和正常组分别给予等量的饮用水,于8周、12周末两个时点处死大鼠。测定血清丙氨酸氨基转移酶(ALT)、门冬氨酸氨基转移酶(AST)、总胆固醇(TC)、甘油三酯(TG);测定空腹血糖(FBG)、空腹胰岛素(FINS),计算胰岛素敏感指数(ISI);HE染色观察肝组织病理变化;RT-PCR分别检测各组大鼠肝脏L-FABP的表达。结果:模型组大鼠血清ALT、AST、TC、TG升高,ISI降低,伴典型的脂肪性肝炎;与模型组相比,治疗组的各项指标比同期模型组都有所改善,两组间有统计学差异。模型组大鼠肝组织的L-FABP mRNA表达增高,与模型组相比治疗组L-FABP mRNA表达显著降低,说明降脂益肝冲剂能减少脂肪肝大鼠肝组织L-FABP mRNA的表达。结论:降脂益肝冲剂能通过调节L-FABP的表达来调节脂质代谢,改善胰岛素抵抗,降低肝脏的脂质沉积,有效地治疗大鼠非酒精性脂肪肝。
Objective: To observe the effect of Jiang Zhi Yi Gan Granules on hepatic fatty acid-binding protein (L-FABP) mRNA expression in non-alcoholic fatty liver (NAFLD) rats. METHODS: Sixty male Wistar rats were randomly divided into 6 groups (10 in each group): Normal group (N1, N2) was given normal feed; Model group (M1, M2) and treatment group (D1, D2) were given high fat. Dietary feeding; D1 group was given the lipid-lowering and Yigan granules at 0 weeks on the basis of the high-fat diet; D2 group was given the lipid-lowering and Yigan granules at the 8th week, while the model group and the normal group were given the same amount of drinking. In water, the rats were sacrificed at both the 8th and 12th weekends. Determination of serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), total cholesterol (TC), triglyceride (TG); determination of fasting blood glucose (FBG), fasting insulin (FINS), calculation Insulin sensitivity index (ISI); hematoxylin-eosin (HE) staining was used to observe the pathological changes of liver tissue; RT-PCR was used to detect the expression of liver L-FABP in each group. RESULTS: Serum ALT, AST, TC, and TG levels in the model group increased, and ISI decreased with typical steatohepatitis. Compared with the model group, each index of the treatment group improved compared with the model group at the same time. There are statistical differences. The expression of L-FABP mRNA in liver tissue of model rats was increased. Compared with the model group, the expression of L-FABP mRNA was significantly decreased in the treatment group, indicating that Jiangzhi Yigan Granule can reduce the expression of L-FABP mRNA in fatty liver rat liver tissue. Conclusion: Jiangzhi Yigan Granule can regulate the expression of L-FABP to regulate lipid metabolism, improve insulin resistance, reduce lipid deposition in the liver, and effectively treat non-alcoholic fatty liver in rats.
