目的:研究家蝇幼虫乙醇提取物(MDE)对酵母致炎发热大鼠的解热作用及机制。方法:利用干酵母致炎大鼠发热动物模型,设立生理盐水组:ig等体积生理盐水;模型组:ig等体积生理盐水+大鼠背部sc 15%干酵母20 mL.kg-1;阿司匹林组:ig阿司匹林0.3 g.kg-1;给药组:分别ig MDE(1,0.5,0.25 g.kg-1),qd,预防性连续给药7 d(n=10),观察MDE对发热大鼠直肠温度的影响,测定给药后不同时间的体温变化,并采用放免法测定下丘脑中前列腺素E2(PGE2)和环磷腺苷(cAMP)含量。结果:1,0.5,0.25 g.kg-1MDE具有不同程度的退热作用;并能减少致热大鼠下丘脑中PGE2及cAMP的含量,高剂量组(126.86±24.89)ng.g-1,(9.72±1.11)nmol.g-1;中剂量组(180.12±34.58)ng.g-1,(10.05±0.26)nmol.g-1;低剂量组(203.54±30.46)ng.g-1,(10.79±0.57)nmol.g-1;与模型组(253.70±43.96)ng.g-1,(12.15±3.11)nmol.g-1比较具有显著性差异(P<0.01,P<0.05)。结论:MDE具有解热作用,其作用机制与降低下丘脑中PGE2以及cAMP有关。
Objective: To study the antipyretic effect and mechanism of ethanol extract of Musca domestica larvae on inflammatory and pyrogenic yeast in rats. Methods: The animal model of fever induced by dry yeast in rats was established. The normal saline group: ig equal volume of normal saline; the model group: ig equal volume of normal saline + rat 15% dry yeast sc 15% dry yeast; aspirin group : ig aspirin 0.3 g.kg-1; administration group: ig MDE (1, 0.5, 0.25 g.kg-1), qd, prophylactic continuous administration for 7 days The effects of different doses of PGE2 and cAMP on the hypothalamus were determined by radioimmunoassay. Results: 1, 0.5, 0.25 g.kg-1MDE had different degrees of antipyretic effect, and could reduce the content of PGE2 and cAMP in the hypothalamus of pyrogen-induced rats. The high dose group (126.86 ± 24.89) ng.g- (9.72 ± 1.11) nmol.g-1, middle dose group (180.12 ± 34.58) ng.g-1, (10.05 ± 0.26) nmol.g-1 and low dose group (203.54 ± 30.46) ng.g- (10.79 ± 0.57) nmol.g-1, which was significantly lower than that of the model group (253.70 ± 43.96) ng.g-1 and (12.15 ± 3.11) nmol.g-1 (P <0.01, P <0.05). Conclusion: MDE has antipyretic effect and its mechanism is related to decreasing PGE2 and cAMP in hypothalamus.
