目的观察绿色荧光蛋白(green fluorescent protein,GFP)转基因鼠来源的胚胎神经上皮干细胞体外增殖和分化的情况。方法将来源于孕11dGFP转基因鼠的胚胎神经上皮干细胞与普通大鼠神经上皮干细胞进行体外培养,比较二者克隆增殖、分化的状况。结果GFP转基因鼠胚胎神经上皮干细胞可在体外进行克隆增殖,并能分化为神经元和神经胶质细胞,与普通鼠神经上皮干细胞比较没有差异。结论GFP转基因鼠的胚胎神经上皮干细胞可在体外克隆及分化,并能稳定地表达绿色荧光蛋白。
Objective To observe the proliferation and differentiation of embryonic neural epithelial stem cells derived from green fluorescent protein (GFP) transgenic mice in vitro. Methods Embryonic neural epithelial stem cells derived from pregnant 11dGFP transgenic mice and normal rat neural epithelial stem cells were cultured in vitro. The proliferation and differentiation of the two clones were compared. Results GFP transgenic mouse embryonic neural epithelial stem cells could proliferate in vitro and differentiate into neurons and glial cells, which showed no difference with normal mouse neural epithelial stem cells. Conclusion GFP transgenic mouse embryonic neural epithelial stem cells can be cloned and differentiated in vitro and stably express green fluorescent protein.
