目的 制备出适合于组织工程用的温固化型水凝胶 ,并将其应用于可注射软骨的建造 ,观察实验结果 .方法 制备不同浓度的水凝胶 ,并添加不同的物质 ,测量其凝胶化温度 ,并把水凝胶用于裸鼠组织工程软骨的建造 .注射后 4,8和 12 wk取材 ,通过大体标本和组织学观察研究软骨的形成情况 .结果 2 5 0 g· L- 1的水凝胶具有恰当的凝胶化温度和能满足要求的力学性能 .含钙磷的添加物因溶解度不高 ,对凝胶的性能未能产生显著影响 .此水凝胶用于软骨组织工程 ,注射后 4wk在皮下形成稍硬的结节 .8wk后形成的结节质地坚韧 ,切面色泽亮白 .组织学观察 4wk即有软骨形成 ,但不成熟 ,8wk标本有成熟软骨形成 ,12 wk标本成熟软骨的量进一步增加 .结论 温固化型水凝胶凝胶化温度受浓度影响 ,2 5 0 g· L- 1 水凝胶凝胶化温度为 5℃ ,可用于组织工程软骨的建造 ,并能顺利长出软骨
