目的探讨胃癌组织中细胞周期素G1(cyclinG1)和cyclinG2基因的表达及其意义。方法采用逆转录聚合酶链反应检测了55例胃癌组织、正常胃组织中的cyclinG1和cyclinG2基因的mRNA表达,流式细胞术检测cyclinG1蛋白的细胞周期分布和表达水平,对cyclinG基因进行测序分析。结果胃癌组织中cyclinG1mRNA表达的阳性率为58%,明显高于正常胃组织(18%,P<0.05),cyclinG1mRNA与蛋白表达之间有正相关性(r=0.456,P=0.000);胃癌组织的cyclinG1蛋白表达阳性率为74.3%,正常胃组织为2.3%,二者之间差异有统计学意义(P<0.05)。CyclinG1蛋白表达于G0/G1期开始出现,在G2/M期达到高峰,有高峰延迟现象。CyclinG2mRNA在胃癌组织中的表达水平为0.5±0.5,正常胃组织中为2.0±0.4,两者之间差异有统计学意义(P<0.01)。CyclinG1与cyclinG2之间表达无相关性(r=-0.217,P=0.063)。CyclinG基因测序结果与genebank中cDNA序列完全一致,没有发现突变点、缺失点或移位点。结论CyclinG的异常表达可能与胃癌的恶性程度和发生发展有关。
Objective To investigate the expression of cyclin G1 and cyclinG2 in gastric cancer and its significance. Methods The mRNA expressions of cyclinG1 and cyclinG2 in 55 gastric cancer tissues and normal gastric tissues were detected by reverse transcription polymerase chain reaction (RT - PCR). The cell cycle distribution and expression of cyclinG1 were detected by flow cytometry. The cyclinG gene was sequenced. Results The positive rate of cyclin G1 mRNA expression in gastric cancer was 58%, which was significantly higher than that in normal gastric tissue (18%, P <0.05). There was a positive correlation between the expression of cyclin G1 mRNA and protein (r = 0.456, P = 0.000) The positive rate of cyclinG1 protein was 74.3% and normal gastric tissue was 2.3%, the difference was statistically significant (P <0.05). The expression of Cyclin G1 protein began to appear in G0 / G1 phase and peaked in G2 / M phase with peak delay. The expression level of CyclinG2 mRNA in gastric cancer tissue was 0.5 ± 0.5, and in normal gastric tissue was 2.0 ± 0.4, the difference was statistically significant (P <0.01). There was no correlation between CyclinG1 and cyclinG2 (r = -0.217, P = 0.063). CyclinG gene sequencing and genebank cDNA sequence exactly the same, found no mutation point, deletion or shift point. Conclusion The abnormal expression of CyclinG may be related to the malignant degree and occurrence and development of gastric cancer.
