本文报告从一例食管鳞癌患者手术切除物边缘剪取一小块组织,做组织块培养。当长出大量成纤维细胞及上皮细胞时,用胰酶-EDTA选择性地将成纤维细胞消化下来,接种到新的培养瓶内,并分成对照组和实验组。实验组用化学致癌物N-甲基-N’-硝基-N-亚硝基胍(简称MNNG)处理。24小时后,用新鲜培养液洗三次并更换新的生长液,继续培养约12周后出现转化灶,可在瓶中无限生长。此转化细胞定名为HEFl。不加致癌物的对照组,传至第三代便全部退化死亡。
This article reports from a case of esophageal squamous cell carcinoma in patients with surgical margins cut a small piece of tissue, tissue culture. When a large number of fibroblasts and epithelial cells were grown, fibroblasts were selectively digested with trypsin-EDTA and inoculated into new flasks and divided into control and experimental groups. The experimental group was treated with the chemical carcinogen N-methyl-N’-nitro-N-nitrosoguanidine (MNNG). After 24 hours, wash with fresh medium three times and change to a new growth solution. Continue culturing for about 12 weeks. Foci appear and grow infinitely in the bottle. This transformed cell is named HEF1. Carcinogen-free control group, passed to the third generation will all degenerate and die.
