目的:建立盐酸美沙酮浓度的快速高效液相色谱法测定方法。方法:以盐酸氯胺酮为内标,血清样品通过硫酸锌(0.005g)沉淀,采用ODS C18(250mm×4.6mm,5μm)色谱分析柱,以乙腈-水-三乙胺(65∶35∶0.3)为流动相,流速为1.0mL.min-1,检测波长为292nm,柱温为(25±1)℃,进样量为20μL。结果:血清样品在0.05~1.00mg.L-1的范围内呈良好的线性关系,r=0.999 1。日内、日间的相对标准偏差RSD均<5%,平均回收率为102.8%。结论:本法简便、快速、准确。
Objective: To establish a method for the determination of methadone hydrochloride by high performance liquid chromatography. METHODS: Ketamine hydrochloride was used as an internal standard. Serum samples were precipitated by zinc sulfate (0.005 g) and eluted with acetonitrile-water-triethylamine (65:35:0.3) using ODS C18 (250 mm × 4.6 mm, As mobile phase, the flow rate was 1.0mL.min-1, the detection wavelength was 292nm, the column temperature was (25 ± 1) ℃, and the injection volume was 20μL. Results: Serum samples showed good linearity in the range of 0.05-1.00 mg.L-1, r = 0.999 1. The relative standard deviations (RSDs) were both less than 5% and the average recovery was 102.8%. Conclusion: This method is simple, fast and accurate.
