麦迪霉素的效价一般用生物测定法测定,结果准确但测定时间长,尤其不能满足生产中发酵液效价测定对时间上的要求。本文所述化学效价测定法经在生产中使用,结果基本与生物效价法相符。 试验运用麦迪霉素与27N硫酸作用水解生成紫色产物,其颜色深浅与麦迪霉素强度之间关系符合比耳定律的原理,采用分光光度法测定。样品为四川抗菌素工业研究所提供的标准品及四川长征制药厂试验室生产的麦迪霉素。测定用72型分光光度计。
The titer of midecamycin is usually determined by bioassay, the result is accurate but the measuring time is long, especially it can not meet the time requirement of determination of titer of broth in the production. The chemical titer method described in this article is used in the production, the results are basically consistent with the biological potency method. Experimental use of mithramycin and 27N sulfuric acid hydrolysis generated purple product, the color depth and the relationship between the intensity of midecamycin in line with Beer’s law principle, determined by spectrophotometry. Samples for the Sichuan Institute of Antibiotics standards and laboratories produced in Sichuan Changzheng Midecamycin. Determination with 72 spectrophotometer.
