目的:建立制备免疫刺激复合物(ISCOMs)的方法,ISCOMs是一种疫苗制备新技术,具有突出的免疫学价值。方法:本研究以人巨细胞病毒(HCMV)包膜蛋白和皂苷类佐剂QuilA作用,采用非离子去垢剂Mega-10及超速离心技术,进行了HCMVISCOMs实验型疫苗的制备。结果:浓缩样品液经蔗糖梯度超速离心后,在离心管底部可见白色病毒带;纯化样品电镜观察具有典型的ISCOMs形态。结论:本研究制备样品在电镜下观察见抗原蛋白分布在ISCOMs颗粒表面,为直径30nm左右的球状结构,与国外文献所报道形态相符,证实了制备的成功
OBJECTIVE: To establish a method for preparing immunostimulating complexes (ISCOMs). ISCOMs is a new vaccine preparation technology with outstanding immunological value. Methods: In this study, HCMV envelope protein and saponin adjuvant QuilA role, the use of non-ionic detergent Mega-10 and ultracentrifugation technology, HCMVISCOMs experimental vaccine preparation. Results: After the supernatant was sucrose gradient sucrose gradient ultracentrifugation, the white virus band was visible at the bottom of the centrifuge tube. The typical ISCOMs morphology was observed by electron microscopy. CONCLUSION: The prepared sample in this study shows that the distribution of antigen protein on the surface of ISCOMs particles under electron microscope is a globular structure with a diameter of about 30nm, which is consistent with the reported morphology in foreign literature, confirming the success of preparation
