目的:观察人参总皂苷与三七总皂苷对高糖刺激下腹膜间皮细胞分泌转化生长因子β1(TGF-β1)和纤维连接蛋白(FN)的影响。方法:采用胰蛋白酶消化法分离大鼠腹膜间皮细胞,建立稳定的细胞培养模型,加入2.5%腹膜透析液作用3h,吸弃后加入终浓度为100μg/mL的人参总皂苷与三七总皂苷,分别于24h、48h后用ELISA法检测上清液中TGF-β1和FN的水平。结果:高糖腹透液刺激后,在24h、48h腹膜间皮细胞分泌FN及TGF-β1均明显增加,与空白组比较,差异有显著性意义或非常显著性意义(P<0.05,P<0.01)。对高糖刺激后的腹膜间皮细胞培养24h,与高糖组比较,三七总皂苷能抑制FN分泌(P<0.01);人参总皂苷培养24h、48h均能降低FN的水平(P<0.01)。人参总皂苷培养24h能减少高糖刺激后腹膜间皮细胞分泌TGF-β1(P<0.01),三七总皂苷培养24h、48h均能降低TGF-β1的水平(P<0.05,P<0.01)。结论:三七总皂苷及人参总皂苷对高糖培养24h的腹膜间皮细胞分泌FN、TGF-β1均有明显的抑制作用,人参总皂苷对FN分泌的抑制持续时间更长,三七总皂苷对TGF-β1分泌的抑制持续时间更长。
Objective: To observe the effects of ginseng total saponins and total saponins of Panax notoginseng on the secretion of transforming growth factor β1 (TGF-β1) and fibronectin (FN) in peritoneal mesothelial cells under high glucose. Methods: Rat peritoneal mesothelial cells were isolated by trypsin digestion and established a stable cell culture model. The cells were treated with 2.5% peritoneal dialysis solution for 3h. After aspiration, the total ginsenosides and total saponins of Panax notoginseng The levels of TGF-β1 and FN in the supernatant were detected by ELISA at 24h and 48h respectively. Results: After high glucose and dialysate stimulation, the secretion of FN and TGF-β1 in peritoneal mesothelial cells significantly increased at 24h and 48h, respectively. Compared with the blank group, the difference was significant or significant (P <0.05, P < 0.01). Compared with high glucose group, Panax notoginseng saponins could inhibit the secretion of FN (P <0.01), while the total ginsenoside could reduce the level of FN 24 h and 48 h (P <0.01) ). Ginseng total saponin could reduce the secretion of TGF-β1 by peritoneal mesothelial cells (P <0.01) 24h after hypoglycemic stimulation, and reduce the level of TGF-β1 (P <0.05, P <0.01) . Conclusion: The total saponins of Panax notoginseng and total ginsenosides have obvious inhibitory effect on the secretion of FN and TGF-β1 in peritoneal mesothelial cells cultured with high glucose for 24h. The inhibition of the secretion of FN by ginseng saponins lasts longer. The total saponins of Panax notoginseng Inhibition of TGF-β1 secretion lasts longer.
