目的确定病毒 IBDV(infection bursal diseases virus, IBDV)的中和位点,为生产病毒疫苗奠定基础。方法利用噬菌体肽库技术,模拟病毒的病原决定簇,同抗IBDV的单抗相结合,通过对肽库的筛选和测序,从而确定病毒的中和位点的氨基酸序列。结果与IBDV抗原的原序列进行对应查找,发现原序列上的198~208(Asp-Ser-Asp-Arg-Pro-Arg-Val-Tyr-Thr-Ile)这11个氨基酸能够体现中和位点的特点,并且与所测序列有4~5个氨基酸残基的重复。结论该序列就是抗原的中和位点。
Objective To determine the neutralization sites of IBDV and lay the foundation for the production of virus vaccine. Methods The phage peptide library technique was used to mimic the pathogenic determinants of the virus and bind to the monoclonal antibody against IBDV. The amino acid sequence of the virus neutralization site was determined by screening and sequencing of the peptide library. The results showed that the 11 amino acids of 198 ~ 208 (Asp-Ser-Asp-Arg-Pro-Arg-Val-Tyr-Thr-Ile) Of the characteristics, and with the measured sequence of 4 to 5 amino acid residues repeat. Conclusion This sequence is the antigen neutralization site.
