目的:探讨四妙勇安汤及其单体绿原酸、阿魏酸对于凝血酶处理的内皮细胞P-选择素及NF-κb p65mRNA表达的影响。方法:MTT法检测绿原酸、阿魏酸、四妙勇安汤对于内皮细胞活力的影响;0.2 U/mL凝血酶处理内皮细胞15 min作为模型组,三种药物预处理内皮细胞24 h作为干预组,流式细胞术、Elisa检测内皮细胞P-选择素的表达,RT-PCR检测NF-κb p65mRNA的表达。结果:绿原酸102ng/m L、阿魏酸104ng/mL时OD值最大;模型组P-选择素及NF-κb p65mRNA的表达均高于空白组(P<0.05),各干预组P-选择素及NF-κb p65mRNA的表达均低于模型组,组间无差异,但四妙组数值低于其余两组。结论:四妙勇安汤及其单体能减少内皮细胞P-选择素的表达,这一作用可能是通过抑制NF-κb p65mRNA的表达来完成的。
Objective: To investigate the effects of Simiao Yongan Decoction and its monomers chlorogenic acid and ferulic acid on the expression of P-selectin and NF-κB p65mRNA in thrombin-treated endothelial cells. Methods: MTT assay was used to detect the effect of chlorogenic acid, ferulic acid and Simiao Yongan Decoction on endothelial cell viability. Endothelial cells were treated with 0.2 U / mL thrombin for 15 min as model group, and the three drugs pretreated endothelial cells for 24 h The expression of P-selectin in endothelial cells was detected by flow cytometry and Elisa assay. The expression of NF-κB p65 mRNA was detected by RT-PCR. Results: The OD values of chlorogenic acid at 102 ng / mL and ferulic acid at 104 ng / mL were the highest. The expressions of P-selectin and NF-κB p65 mRNA in model group were higher than those in blank group (P <0.05) The expressions of selectin and NF-κB p65mRNA were lower in the model group than in the model group, but there was no difference between the two groups. Conclusion: Simiao Yongan Decoction and its monomer can reduce the expression of P-selectin in endothelial cells, which may be accomplished by inhibiting the expression of NF-κB p65mRNA.
