目的:探讨抑癌基因RIZ1在卵巢上皮性肿瘤组织及卵巢癌细胞系中的表达及其表达变化与卵巢癌发生的关系。方法:以卵巢肿瘤组织及卵巢癌细胞系为研究对象。RT-PCR检测RIZ1 mRNA水平;蛋白印迹检测RIZ1蛋白表达。结果:卵巢癌组织、卵巢癌细胞系、良性肿瘤组织及正常卵巢组织中RIZ1 mRNA相对含量的平均值分别为0.42±0.16、0.34±0.12、0.68±0.12及1.17±0.08,RIZ1蛋白相对含量的平均值分别为0.48±0.11、0.38±0.08、0.82±0.11及1.56±0.14。卵巢癌组织、卵巢癌细胞系、良性卵巢肿瘤组织分别与正常卵巢组织比较,RIZ1基因的表达差异均有统计学意义(P<0.05);良性卵巢肿瘤组织与卵巢癌组织比较,RIZ1基因的表达差异有统计学意义(P<0.05)。结论:RIZ1基因表达缺陷与上皮性卵巢癌的发生密切相关。
Objective: To investigate the expression of tumor suppressor gene RIZ1 in ovarian epithelial tumor and ovarian cancer cell lines and its relationship with the occurrence of ovarian cancer. Methods: The ovarian tumor tissue and ovarian cancer cell lines as the research object. RIZ1 mRNA was detected by RT-PCR and RIZ1 protein was detected by Western blotting. Results: The average relative content of RIZ1 mRNA in ovarian cancer tissue, ovarian cancer cell line, benign tumor tissue and normal ovary tissue were 0.42 ± 0.16,0.34 ± 0.12,0.68 ± 0.12 and 1.17 ± 0.08 respectively. The average relative content of RIZ1 protein The values were 0.48 ± 0.11, 0.38 ± 0.08, 0.82 ± 0.11 and 1.56 ± 0.14, respectively. The expression of RIZ1 gene in ovarian cancer tissue, ovarian cancer cell line and benign ovarian tumor tissue was significantly higher than that in normal ovarian tissue respectively (P <0.05). Compared with ovarian cancer tissue, the expression of RIZ1 gene The difference was statistically significant (P <0.05). Conclusion: The deficiency of RIZ1 gene expression is closely related to the occurrence of epithelial ovarian cancer.
