目的:评价锦南复方对高脂饮食联合链脲佐菌素诱导的2型糖尿病大鼠胰岛素抵抗及葡萄糖转运体4(GLUT-4)、胰岛素受体底物1(IRS-1)表达的影响。方法:高脂饮食联合小剂量链脲佐菌素建立2型糖尿病大鼠模型,观察锦南复方对2型糖尿病大鼠血糖、血脂、抗氧化及胰岛素抵抗指数的影响,使用免疫组化法测肝组织中GLUT-4、IRS-1的表达水平。结果:锦南复方能显著降低糖尿病大鼠餐后血糖(PBG)、空腹血糖(FBG)、糖化血红蛋白(Hb A1c)、甘油三酯(TG)、低密度脂蛋白(LDL-C)、总胆固醇(TC)、游离脂肪酸(FFA)、空腹血清胰岛素(FINS)及丙二醛(MDA)水平和胰岛素抵抗指数(IRI),上调肝糖原含量及肝组织GLUT-4、IRS-1的表达水平,有效改善大鼠的胰岛素抵抗。结论:锦南复方对2型糖尿病大鼠降糖、调脂作用显著,能有效改善糖尿病大鼠胰岛素抵抗,其作用机制可能与上调肝组织中GLUT-4、IRS-1蛋白表达有关。
Objective: To evaluate the effect of Jinnan Compound on insulin resistance and the expression of glucose transporter 4 (GLUT-4) and insulin receptor substrate 1 (IRS-1) in type 2 diabetic rats induced by high-fat diet combined with streptozotocin . Methods: A high-fat diet combined with low-dose streptozotocin was used to establish type 2 diabetic rat model. The effects of Jinnan Compound on blood glucose, blood lipid, antioxidant and insulin resistance index in type 2 diabetic rats were observed. Immunohistochemical method The expression of GLUT-4, IRS-1 in liver tissue. Results: Jinnan Compound can significantly reduce the postprandial blood glucose (PBG), fasting blood glucose (FBG), Hb A1c, triglyceride (TG), low density lipoprotein (LDL-C), total cholesterol (TC), free fatty acid (FFA), fasting serum insulin (FINS), malondialdehyde (MDA) and insulin resistance index (IRI), upregulated the content of hepatic glycogen and the expression of GLUT-4 and IRS- , Effectively improve insulin resistance in rats. CONCLUSION: Jinnan Compound has significant effects on hypoglycemic and lipid-lowering in type 2 diabetic rats and can effectively improve insulin resistance in diabetic rats. Its mechanism may be related to up-regulating the expression of GLUT-4 and IRS-1 in liver tissue.
