环磷酰胺对小鼠精子胞内钙离子浓度的影响

目的观察腹腔注射环磷酰胺对小鼠精子胞内钙离子浓度的影响。方法将20只雄性昆明小鼠随机分为对照组和模型组,按60mg/kg体重给对照组小鼠腹腔注射生理盐水(NS),另按60mg/kg体重给模型组小鼠腹腔注射环磷酰胺,每天1次,连续5d,常规饲养观察34d,然后对小鼠精子进行精液常规分析,对精子行FITC-AV/PI染色后用流式细胞仪检测精子凋亡率,用Ca~(2+)荧光探针Fluo-3/AM处理精子后用流式细胞仪检测精子胞内Ca~(2+)浓度。结果对照组与模型组精子密度分别为(5.20±1.34)×10~6/ml和(1.73±0.03)×10~6/ml,精子活力分别为(14.49±0.30)%和(6.64±1.88)%,精子活率分别为(68.39±1 5.1 3)%和(39.96±4.89)%,精子凋亡率(AV/PI)分别为(8.38±0.45)和(14.84±1.22),胞内Ca~(2+)浓度分别为(133.14±11.86)和(80.84±6.92),以上项目两组比较差异均有统计学意义(P<0.05)。结论腹腔注射环磷酰胺可使小鼠精子密度、活力、活率降低,精子凋亡增加,精子胞内Ca~(2+)浓度降低。造成精子胞内Ca~(2+)浓度降低的可能机理是环磷酰胺降低了精子特异性Ca~(2+)通道蛋白功能。 Objective To observe the effect of intraperitoneal injection of cyclophosphamide on the intracellular calcium concentration of sperm in mice. (2+) concentrations were (133.14 ± 11.86) and (80.84 ± 6.92) respectively. There were significant differences between the two groups in the above items (P <0.05). Conclusion Intraperitoneal injection of cyclophosphamide can reduce the density, viability and viability of sperm, increase sperm apoptosis and decrease intracellular Ca 2+ concentration in sperm. The possible mechanism of the decrease of intracellular Ca2 + concentration in sperm is that cyclophosphamide reduces sperm-specific Ca2 + channel protein function.
