采用改良的PCRRFLP方法测定长沙地区291名儿童载脂蛋白(Apo)E基因型。结果发现ε2,ε3,ε4等位基因的频率分别是0081,0857和0062。ApoE等位基因频率分布无明显的性别差异(P>0.05)。Apoε2等位基因携带者与其它ApoE基因携带者相比,血高密度脂蛋白胆固醇、ApoE和ApoA1浓度较高,而总胆固醇、低密度脂蛋白胆固醇、ApoB100水平较低(P<0.05)。ε4等位基因携带者的血脂、脂蛋白、Apo改变则相反(P<0.05)。结果还显示ApoE等位基因对血甘油三酯、脂蛋白(a)浓度的影响不明显(P>0.05)。ε2,ε3,ε4等位基因与血总胆固醇、低密度脂蛋白胆固醇、ApoB100浓度呈等级正相关,与高密度脂蛋白胆固醇、ApoA1及ApoE水平呈等级负相关(P<0.05)。ApoE基因对血清脂类的影响存在性别差异。ApoE基因多态性影响小儿血清脂类代谢。
The PCR-RFLP method was used to determine the Apo E genotype of 291 children in Changsha. The results showed that the frequencies of ε2, ε3 and ε4 alleles were 0081, 0857 and 0062, respectively. ApoE allele frequency distribution no significant gender differences (P> 0.05). Compared with carriers of other ApoE gene carriers, carriers of Apoε2 allele had higher concentrations of serum high-density lipoprotein cholesterol, ApoE and ApoA1, and lower levels of total cholesterol, LDL-C and ApoB100 (P <0. 05). ε4 allele carriers of blood lipids, lipoprotein, Apo change the contrary (P <0.05). The results also showed that ApoE allele of blood triglyceride, lipoprotein (a) concentration was not significant (P> 0.05). The levels of ε2, ε3 and ε4 alleles were positively correlated with total cholesterol, LDL cholesterol and ApoB100, and negatively correlated with high density lipoprotein cholesterol, ApoA1 and ApoE levels (P0.05) . There are gender differences in the effects of ApoE on serum lipids. ApoE gene polymorphism affects pediatric serum lipid metabolism.
