目的:采用Box-Behnken效应面法,优选闪式提取金银花中有效成分的工艺条件。方法:采用闪式提取法,以乙醇浓度、料液比和提取时间为主要影响因素,以绿原酸和木犀草苷转移率的总评“归一值”为评价指标进行Box-Behnken试验设计,并用多元线性回归及二项式拟合,建立总评归一值与自变量之间的数学关系,用效应面法预测最佳工艺条件。结果:采用闪式提取,乙醇浓度为66.92%,料液比为18.82,提取时间为1.20 min,提取次数为一次。以此工艺条件提取金银花中绿原酸的转移率为92.87%,木犀草苷的得率为87.55%。结论:优选的提取工艺简单、经济实用,能为金银花药材中有效成分的快速提取及质量控制提供新思路。
Objective: The Box-Behnken effect surface method, preferably flash-type extraction of honeysuckle in the active ingredients of the process conditions. Methods: Box-Behnken test was carried out by flash extraction with ethanol concentration, solid-to-liquid ratio and extraction time as the main influencing factors, and the overall rating of “conversion of chlorogenic acid and luteolin” Design and use multivariate linear regression and binomial fit to establish the mathematical relationship between the value and the independent variable of the total assessment, and predict the optimum technological conditions by the effect surface method. Results: By flash extraction, the ethanol concentration was 66.92%, the ratio of material to liquid was 18.82, the extraction time was 1.20 min and the number of extraction was once. The extraction rate of chlorogenic acid in honeysuckle was 92.87% and the yield of luteolin was 87.55%. Conclusion: The optimized extraction process is simple, economical and practical, which can provide new ideas for the rapid extraction and quality control of active ingredients in honeysuckle.
