目的:探讨干扰素α(IFN-α)对膀胱癌细胞多药耐药性的逆转作用。方法:以人膀胱移行上皮癌细胞系BIU87和BIU87/ADM为研究对象,应用MTT法检测阿霉素(ADM)的细胞杀伤作用、荧光法检测细胞内ADM浓度和斑点杂交法检测mdrlmRNA相对含量。结果:500IU/mlIFN-α能明显增强ADM对耐药细胞BIU87/ADM的杀伤作用,但不能增强ADM对敏感细胞BIU87的杀伤作用;IFN-α能增加ADM在BIU87/ADM细胞内的积聚,但并不改变mdrlmRNA在细胞内的表达量。结论:IFN-α对膀胱癌细胞多药耐药性具有逆转作用,其机制与IFN-α增加细胞内抗癌药物浓度有关,与mdrlmRNA表达量无关
Objective: To investigate the reversal effect of interferon alfa (IFN-α) on multidrug resistance of bladder cancer cells. METHODS: Human bladder transitional cell carcinoma cell line BIU87 and BIU87/ADM were studied. MTT assay was used to detect the cytotoxicity of adriamycin (ADM), fluorescence was used to detect intracellular ADM concentration, and blot hybridization was used to detect the relative content of mdrl mRNA. RESULTS: IFN-α at 500 IU/ml could significantly enhance the killing effect of ADM on BIU87/ADM cells, but it could not enhance the killing effect of ADM on BIU87 cells. IFN-α increased the accumulation of ADM in BIU87/ADM cells. Does not change the expression of mdrl mRNA in cells. Conclusion: IFN-α has a reversal effect on the multidrug resistance of bladder cancer cells, and its mechanism is related to the increase of intracellular anticancer drug concentration by IFN-α and not related to the expression of mdrl mRNA.
