目的:运用近红外光谱技术(NIRS)测定不同产地野菊花总黄酮含量。方法:分别采集6个不同产地的野菊花样品,采用近红外漫反射光谱法采集不同产地野菊花近红外图谱;以芦丁为对照品,利用传统的分析方法紫外-可见分光光度法在502 nm处测定不同产地野菊花药材中总黄酮含量作为参考值,将近红外图谱与野菊花中总黄酮含量值进行关联,采用偏最小二乘法(PLS)建立定量分析模型,并对模型进行验证。结果:所建总黄酮定量分析模型的相关系数(R~2)、校正均方差(RMSEC)、预测均方差(RMSEP)和交叉验证均方差(RMSECV)分别为0.989 27、0.131、0.163和0.292;验证集样品的总黄酮含量近红外预测值的平均相对偏差为2.08%。结论:近红外光谱技术测定快速,操作简单,预测结果准确度高,可用于不同产地野菊花药材中总黄酮含量的快速测定。
Objective: To determine the content of total flavonoids in wild chrysanthemum with near infrared spectroscopy (NIRS). Methods: Six wild chrysanthemum samples were collected from different regions of China. Near-infrared diffuse reflectance spectroscopy was used to collect the near-infrared spectrum of chrysanthemum from different regions. Rutin was used as a reference substance and analyzed by UV-Vis spectrophotometry at 502 nm Determination of total flavonoids content of Radix Chrysanthemum from different areas as reference value, correlation between NIR spectrum and total flavonoid content in Chrysanthemum indicum, using partial least squares (PLS) to establish a quantitative analysis model and validate the model. Results: The correlation coefficients (R ~ 2), RMSEC, RMSEP and RMSECV of the total flavonoids were 0.989 27,0.131,0.163 and 0.292 respectively. The average relative deviation of the total flavonoid content of the validation set samples from the near-infrared predicted value was 2.08%. Conclusion: The technique of near infrared spectroscopy is rapid, easy to operate and has high prediction accuracy. It can be used for the rapid determination of total flavonoids in wild chrysanthemum medicinal materials.
