背景与目的:H IF-1α在许多恶性肿瘤中的过表达与肿瘤的血管形成、侵袭转移及放化疗耐受等密切相关。研究发现,H IF-1α的过表达与突变的P53蛋白存在密切关系。本实验探讨H IF-1α在胃癌中表达的意义及其与P53及P糖蛋白(P-gp)的关系。方法:采用SP免疫组织化学染色法检测74例经病理证实的胃癌组织中的H IF-1α、P53及多药耐药蛋白的表达。结果:在H IF-1α蛋白表达阳性的44例中,P53蛋白表达阳性率为70.45%;在H IF-1α蛋白表达阴性的30例中,P53蛋白表达阳性率为30.0%,两者表达呈正相关(rs=0.327,P<0.01)。在H IF-1α蛋白表达阳性的44例中,P-gp蛋白表达阳性率为61.36%,在H IF-1α蛋白表达阴性的30例中,P-gp蛋白表达阳性率为36.67%,两者表达呈正相关(rs=0.256,P<0.01)。在P53蛋白表达阳性的40例中,P-gp蛋白表达阳性率为65%,在H IF-1α蛋白表达阴性的34例中,P-gp蛋白表达阳性率为35.29%,两者表达呈正相关(rs=0.283,P<0.01)。结论:H IF-1α蛋白过表达为影响胃癌预后的独立因素。胃癌组织中P53突变蛋白具有上调H IF-1α的作用,通过P53/H IF-1α途径可能在P糖蛋白调节及多药耐药过程发生重要作用。
BACKGROUND & OBJECTIVE: Overexpression of HIF-1α in many malignant tumors is closely related to tumor angiogenesis, invasion and metastasis, and chemoradiation tolerance. The study found that HIF-1α overexpression and mutation of P53 protein is closely related. This study was to explore the significance of HIF-1α expression in gastric cancer and its relationship with P53 and P-glycoprotein (P-gp). Methods: SP immunohistochemical staining was used to detect the expression of HIF-1α, P53 and multidrug resistance protein in 74 pathologically confirmed gastric cancer tissues. Results: The positive rate of P53 protein expression was 70.45% in 44 cases with positive HIF-1α protein expression, and 30.0% in 30 cases with negative HIF-1α protein expression, the positive expression rate of P53 protein was positive (Rs = 0.327, P <0.01). The positive expression rate of P-gp protein was 61.36% in 44 cases with positive HIF-1α protein expression, and 36.67% in 30 cases with negative HIF-1α protein expression The expression was positively correlated (rs = 0.256, P <0.01). The positive expression rate of P-gp was 65% in 40 cases with P53 protein positive and 35.29% in 34 cases with negative HIF-1α protein expression, the positive expression of P-gp protein was positive (rs = 0.283, P <0.01). Conclusion: HIF-1α overexpression is an independent factor affecting the prognosis of gastric cancer. P53 mutein in gastric cancer has the effect of up-regulating HIF-1α. P53 / HIF-1α pathway may play an important role in the regulation of P-glycoprotein and multidrug resistance.
