目的观察益智汤对APP695转基因小鼠行为学及脑组织β淀粉样蛋白(Aβ)及其前体蛋白(APP)的影响,探讨益智汤治疗阿尔茨海默病(AD)的机理。方法3月龄APP695转基因小鼠随机分为模型组、益智汤组,同月龄同背景转基因阴性小鼠为正常组。益智汤组给予益智汤0.1 g/kg灌胃,模型组和正常组给予等体积蒸馏水。益智汤治疗3个月后,使用电迷宫检测各组小鼠学习记忆能力,采用免疫组织化学方法观察各组小鼠大脑皮层、海马APP和Aβ的表达情况。结果电迷宫检测发现,模型组小鼠学习记忆能力低于正常组(P<0.05),益智汤组小鼠学习记忆能力较模型组增强(P<0.05)。免疫组织化学结果提示,模型组小鼠脑组织各区APP和Aβ的表达高于正常组(P<0.01),益智汤组小鼠脑组织各区APP和Aβ的表达比模型组明显减弱(P<0.05)。结论APP695转基因小鼠是研究AD的良好模型,益智汤通过下调APP和Aβ的表达来改善其学习记忆能力。
Objective To observe the effect of “Yizhi Decoction” on behavior and brain Aβ and its precursor protein (APP) in APP695 transgenic mice and explore the mechanism of Yizhi Decoction in treating Alzheimer’s disease (AD). Methods Three-month-old APP695 transgenic mice were randomly divided into model group, Yizhi Decoction group and normal transgenic mice with the same age as the background. Yizhi Decoction group was given 0.1g / kg Yizhi Decoction, the model group and normal group were given equal volume of distilled water. Three months after treatment with Yizhi decoction, learning and memory abilities of mice in each group were detected by electrical maze. The expression of APP and Aβ in hippocampus and hippocampus was observed by immunohistochemistry. Results The results of electrical maze test showed that the learning and memory abilities of mice in model group were lower than those in normal group (P <0.05). The learning and memory abilities of Yizhi decoction group were higher than that of model group (P <0.05). The results of immunohistochemistry showed that the expression of APP and Aβ in the model group was higher than that in the normal group (P <0.01), and the expression of APP and Aβ in the brain tissue of the Yizhi decoction group was weaker than the model group (P < 0.05). Conclusion APP695 transgenic mice is a good model for studying AD. Yizhi decoction can improve learning and memory by down-regulating the expression of APP and Aβ.
