目的:建立高效液色谱法同时测定管花肉苁蓉中主要药效成分(松果菊苷、麦角甾苷)含量的方法。分析比较2个寄主,4个产地,4批次的管花肉苁蓉中主要药效成分的含量,为合理利用开发管花肉苁蓉提供科学依据。方法:采用HPLC法,色谱柱:Kromasil C_(18)(4.6 mm×250 mm,5μm);流动相:甲醇-乙腈-1%乙酸溶液(14∶9∶77);检测波长:334 nm;流速:1.0 mL·min-1;柱温30℃;结果:2种成分的标准曲线在检测范围内呈良好线性(r=0.999,9)。加样回收率平均值为96.89%、100.7%,RSD%为0.91%与1.96%。分析结果表明,不同产地、批次、寄主的管花肉苁蓉,其主要药效成分(松果菊苷、麦角甾苷)的含量明显不同。结论:该方法简便、准确,重复性好。其所测数据为管花肉苁蓉的药用价值提供实验依据。
Objective: To establish a method for the simultaneous determination of the main active ingredient (echinacoside and ergosteroid) in Cistanche tubulosa by high performance liquid chromatography. Analysis and comparison of the contents of the main active ingredients in the Cistanche tubulosa from two host plants, four producing areas and four batches provide scientific basis for the rational utilization of the Cistanche tubulosa. Methods: The HPLC method was adopted. The mobile phase consisted of Kromasil C 18 (4.6 mm × 250 mm, 5 μm), mobile phase methanol-acetonitrile 1% acetic acid solution (14:9:77) : 1.0 mL · min-1; column temperature was 30 ℃; Results: The standard curve of the two components showed a good linearity (r = 0.999,9) within the detection range. The average recovery rate was 96.89%, 100.7%, RSD% 0.91% and 1.96%. The results showed that the content of erucic acid and echinacoside were significantly different in different regions, batches and hosts of Cistanche tubulosa. Conclusion: The method is simple, accurate and reproducible. The measured data provide the experimental evidence for the medicinal value of Cistanche tubulosa.
