目的探讨胃康宁对细胞内电压依赖性离子通道(VDAC1)的调节和三磷酸腺苷(ATP)水平的干涉效应及其之间的联系。方法体外培养人胃黏膜上皮细胞(GES-1),实验分为空白对照组、阳性对照组(多潘立酮),胃康宁低、中、高剂量组,用逆转录荧光定量聚合酶链反应(RTPCR)和Western blot法检测VDAC1的mRNA和蛋白表达,生物发光法分别检测给药后细胞内能量ATP的含量;制备功能性消化不良大鼠模型,检测胃康宁对功能性消化不良大鼠胃组织中ATP代谢相关的Ca2+-Mg2+-ATP酶的影响。结果 (1)胃康宁在基因和蛋白水平上促进VDAC1的表达和细胞内ATP含量。(2)胃康宁促进功能性消化不良大鼠胃组织Ca2+-Mg2+-ATP酶活性升高。结论VDAC1在基因和蛋白水平上表达增加是胃康宁诱导细胞内能量ATP水平升高的分子机制之一,并且体内实验证明胃康宁促进Ca2+-Mg2+-ATP酶活性,从而对ATP的产生有一定的保护作用,胃康宁作用机制有可能通过能量代谢的途径对功能性消化不良进行治疗。
Objective To investigate the interference effect of Weikangning on the regulation of voltage-dependent ion channel (VDAC1) and the level of adenosine triphosphate (ATP) in cells and its relationship. Methods Human gastric mucosal epithelial cells (GES-1) were cultured in vitro. The experiment was divided into blank control group, positive control group (domperidone) and Weikening low, medium and high dose groups. Reverse transcriptase quantitative polymerase chain reaction And Western blot were used to detect the mRNA and protein expression of VDAC1. The bioluminescence method was used to detect the intracellular energy ATP content after administration. Functional dyspepsia rat model was prepared and gastric perfusion of Weichengling Metabolism Related Ca2 + -Mg2 + -ATP Enzymes. Results (1) Weikangning promoted VDAC1 expression and intracellular ATP content at the gene and protein levels. (2) Weikangning promoted the increase of Ca2 + -Mg2 + -ATPase activity in gastric tissue of rats with functional dyspepsia. Conclusion The increased expression of VDAC1 at the gene and protein level is one of the molecular mechanisms of gastric adenylyltransferase (ATP) -induced intracellular ATP upregulation, and in vivo experiments show that Weikangning can promote the activity of Ca2 + -Mg2 + -ATPase and thus produce certain ATP Protective effect, stomach Corning mechanism of action may be through energy metabolism pathway for functional dyspepsia treatment.
