目的 :探讨苦参素注射液对肿瘤血管形成的抑制作用。方法 :通过建立动物肿瘤模型 ,运用分别实验治疗 ,结合免疫组化 ,检测其抑瘤率和肿瘤内血管密度 ,以及瘤体血管内皮生长因子 (VEGF)和碱性成纤维细胞生长因子 (bFGF)表达的改变。结果 :苦参素具有明显的抑制S180 肉瘤作用 ,其大剂量组抑瘤率为 31.36 %。中、大剂量苦参素组的S180 瘤体内微血管密度均明显低于对照组 ;免疫组化显示苦参素大剂量组可抑制S180 瘤体内VEGF、bFGF的表达。结论 :苦参素对小鼠S180 肉瘤有一定的抑制作用 ,对S180 肉瘤血管形成有明显的抑制作用 ,降低VEGF ,bFGF的表达可能是其抑制肿瘤血管形成的主要机制之一。
Objective: To investigate the inhibitory effect of oxymatrine injection on tumor angiogenesis. Methods: The animal model of tumor was established by using experimental treatment and immunohistochemistry respectively. The tumor inhibition rate and tumor density in the tumor, as well as tumor vascular endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF) Changes in expression. Results: Oxymatrine had a significant inhibitory effect on S180 sarcoma, and its inhibitory rate in high dose group was 31.36%. Medium and high doses of matrine in S180 tumor microvessel density were significantly lower than the control group; immunohistochemistry showed that high dose of matrine can inhibit S180 tumor VEGF, bFGF expression. Conclusion: Oxymatrine can inhibit S180 sarcoma in mice and inhibit the angiogenesis of S180 sarcoma. Decreasing the expression of VEGF and bFGF may be one of the main mechanisms that inhibit the tumor angiogenesis.
