目的探讨胃泌素在人胃癌细胞株(SGC7901)的表达以及17肽胃泌素和抗胃泌素McAb对胃癌细胞株生长的影响。方法以自制的抗胃泌素McAb为一抗,采用免疫组化链亲和素生物素复合物法(streptavidinbiotincomplex,SABC),检测胃泌素在人胃癌细胞株SGC7901的表达,并用17肽胃泌素和抗胃泌素McAb作用于SGC7901细胞株,通过MTT法检测细胞增殖水平。结果胃泌素在SGC7901细胞株呈强阳性表达,17肽胃泌素在1×10-9mol/L~1×10-5mol/L浓度范围内能显著促进该细胞株的生长,抗胃泌素McAb不仅能抑制胃癌细胞的生长,而且还可抑制17肽胃泌素对细胞的促生长作用。结论胃泌素通过自分泌方式促进人胃癌细胞株SGC7901的生长,抗胃泌素McAb能抑制胃癌细胞的生长,以抗胃泌素McAb为基础的肿瘤靶向生物治疗有进一步研究的价值。
Objective To investigate the expression of gastrin in human gastric cancer cell line (SGC7901) and the effects of gastrin 17 and anti-gastrin McAb on the growth of gastric cancer cell lines. Methods Using self-made anti-gastrin McAb as primary antibody, the expression of gastrin in human gastric cancer cell line SGC7901 was detected by immunohistochemical streptavidin biotin complex (SABC) Sucrose and anti-gastrin McAb in SGC7901 cell line, by MTT assay cell proliferation. Results Gastrin expression was strongly positive in SGC7901 cell line. The concentration of 17 gastrin in the concentration range of 1 × 10-9mol / L to 1 × 10-5mol / L could significantly promote the growth of this cell line. McAb can not only inhibit the growth of gastric cancer cells, but also inhibit the role of 17 gastrin on cell growth. Conclusion Gastrin can promote the growth of human gastric cancer cell line SGC7901 through autocrine. Anti-gastrin McAb can inhibit the growth of gastric cancer cells. Further research is worth to be carried out on tumor-targeted biological therapy based on anti-gastrin McAb.
