目的:研究理气散结汤对大鼠乳腺增生的治疗作用,并对其作用机制进行初步探讨。方法:采用肌内注射雌-孕激素联合诱导制备乳腺增生模型大鼠,选择造模成功的大鼠按体质量随机分为模型组、小金胶囊组、理气散结汤低、中、高剂量组,另设正常对照组。连续给药30 d后,测定各组大鼠胸腺、脾脏和子宫重量系数,观察乳腺组织病理形态改变,ELISA法测定血清雌二醇(E2)和黄体酮(P)含量,RT-PCR检测雌激素受体α(ERα)基因表达水平,Western-blot检测ERα蛋白表达水平。结果:与模型组比较,理气散结汤能显著增加乳腺增生大鼠脾脏和胸腺重量指数(P<0.05),减小子宫重量指数(P<0.05),显著减轻大鼠乳腺增生程度,显著降低血清E2含量(P<0.05)、增加P的含量(P<0.05),显著下调ERα基因和单边表达水平(P<0.05)。结论:理气散结汤对大鼠乳腺增生有良好的治疗作用,其机制与调节乳腺增生大鼠血清中性激素及其受体水平有关。
Objective: To study the therapeutic effect of “Liqi Sanjie Decoction” on mammary gland hyperplasia in rats and its mechanism of action. Methods: The model of mammary gland hyperplasia was induced by intramuscular injection of estrogen and progesterone. The successful rats were randomly divided into model group, Xiaojin capsule group, low and medium dose group and high dose group , Another set of normal control group. Thirty days after continuous administration, the thymus, spleen and uterus weights of rats in each group were measured, the pathological changes of breast tissues were observed, the content of estradiol (E2) and progesterone (P) The level of ERα gene expression was detected by Western-blot. Results: Compared with the model group, Liqi Sanjie Decoction could significantly increase the weight index of spleen and thymus (P <0.05), reduce the weight of uterus (P <0.05), significantly reduce the degree of mammary gland hyperplasia and decrease significantly (P <0.05), increase the content of P (P <0.05), and significantly reduce the level of ERα gene and unilateral expression (P <0.05). Conclusion: Liqi Sanjie has a good therapeutic effect on hyperplasia of mammary gland in rats, and its mechanism is related to the regulation of serum hormones and their receptors in mammary gland hyperplasia rats.
