建立了超高效液相色谱-串联质谱法测定Beagle犬血浆中的尼莫地平。以尼群地平为内标,使用Waters Cortecs C18色谱柱,采用电喷雾电离源(ESI),多反应监测(MRM)模式,正离子检测。尼莫地平在0.1~300 ng/ml范围内线性关系良好。日内、日间RSD均小于15%。同时测定了Beagle犬口服尼莫地平片后的血药浓度,主要药动学参数分别为:t_max(0.7±0.5)h,t_1/2(5.5±3.9)h,c_max(322.3±181.0)ng/ml,AUC0→t(833.3±412.7)ng·h·ml-1,AUC0→∞(870.6±448.9)ng·h·ml-1,MRT(5.2±2.3)h。
A ultra performance liquid chromatography-tandem mass spectrometry was established for the determination of nimodipine in Beagle dogs plasma. Nitrendipine was used as an internal standard with a Waters Cortecs C18 column using electrospray ionization (ESI), multiple reaction monitoring (MRM) mode, positive ion detection. Nimodipine has good linearity in the range of 0.1-300 ng / ml. Days, day RSD were less than 15%. The plasma concentrations of nimodipine tablets in Beagle dogs were determined at the same time.
